Retatrutide is a synthetic peptide designed to activate three receptors simultaneously, namely GIPR, GLP 1R, and GCGR. Because it has activity against these three targets, retatrutide is often referred to as a triple agonist or triple hormone receptor agonist.
This approach differs from GLP-1 agonists, which primarily act through GLP-1R, or dual agonists such as tirzepatide, which act on GIPR and GLP-1R. This compound also targets the glucagon receptor, which is expected to provide additional effects on energy regulation and metabolism.

What Are the Potential Side Effects of Retatrutide?
The mechanism of action of this compound is associated with the activation of three hormonal pathways.
1. GLP 1 receptor activation

As a peptide molecule with a relatively complex structure, the selection of an analytical method should be adjusted according to the testing objectives and sample characteristics.
One method that can be used is reverse phase high performance liquid chromatography (RP HPLC). This method can help separate retatrutide from related compounds, impurities, and degradation products based on differences in their interactions with the stationary and mobile phases.
The resulting chromatographic data can then be used to evaluate chromatographic purity and, when the method has been properly developed and validated using an appropriate reference standard, to determine the retatrutide content. In addition to RP HPLC, liquid chromatography mass spectrometry (LC MS) or liquid chromatography high resolution mass spectrometry (LC HRMS) can be used to help confirm the identity of retatrutide.
Mass spectrometry provides information about molecular mass, complementing the data obtained from chromatography. This is important because retention time in HPLC alone may not always be sufficient to definitively confirm the identity of a peptide.
LC MS or LC HRMS can also provide information about related compounds, degradation products, and other molecular characteristics that may be present in the sample.
